Purification and Biochemical Characterization of the DNA Strand-exchange Protein Dmc1 from Fission Yeast

Abstract

DNA strand exchange is a central step of homologous recombination. In meiosis, Dmc1 mediates the strand exchange, which is required for reciprocal crossover between homologous parental chromosomes. The strand exchanging protein forms right helical nucleoprotein filament on single-stranded DNA (ssDNA) to promote strand exchange between the ssDNA and its intact, complementary double-stranded DNA (dsDNA). Several accessory proteins are required to directly control Dmc1’s activity in the reaction. In this section, we shows purification of the fission yeast Dmc1 and its accessory proteins and describes a conventional method to monitor DNA strand exchange reaction, which is a powerful tool to understand the biological significance of Dmc1 as well as its accessory proteins.
